چکیده

The human X chromosome is the favorite chromosome of many geneticists because of its peculiar inheritance pattern and unique biology. The peculiar inheritance pattern stems from its hemizygosity in males and gives rise to a disproportionately high number of Mendelian diseases associated with a human chromosome. As high as 70% of the genes with a known function on the X are associated with disease phenotypes. The unique biology, termed Xinactivation, describes the transcriptional silencing of one of its copies in females. A substantial deal of sex differences in health issues has its origins in the genes on the X chromosomes and the X-inactivation process [1]. For example, a single copy of X chromosome in males leads to their vulnerability to X-linked diseases, such as X-linked immunological diseases (Bruton agammaglobulinemia and Wiskott–Aldrich syndrome), Duchenne muscular dystrophy, or a variety of X-linked mental retardation syndromes, such as fragile X syndrome. Theory and treatment of the X-inactivation chimera in female-prevalent autoimmune disease.

The vast majority of patients with systemic autoimmune diseases such as Sjögren's syndrome and systemic lupus erythematosus are female. The female prevalence of such autoimmune diseases is poorly understood. In theory, X-chromosome inactivation and resultant tissue chimerism may explain the female predisposition to systemic autoimmunity. For example, autoreactive T cells may fail to be tolerize...

X Chromosome Inactivation in Opioid Addicted Women

Introduction: X chromosome inactivation (XCI) is a process during which one of the two X chromosomes in female human is silenced leading to equal gene expression with males who have only one X chromosome. Here we have investigated XCI ratio in females with opioid addiction to see whether XCI skewness in women could be a risk factor for opioid addiction. Methods: 30 adult females meeting DS...

Concurrent FOXP3- and CTLA4-associated genetic predisposition and skewed X chromosome inactivation in an autoimmune disease-prone family.

CLTA4 is relevant for FOXP3(+)Treg cells, and the link between skewed X chromosome inactivation (XCI) and autoimmunity is recognized. The observation of immune dysregulation polyendocrinopathy enteropathy X-linked syndrome and multiorgan endocrine autoimmune phenomena in various members of one family, associated with a CTLA4 polymorphism and skewed XCI, provides an in vivo model of how mechanis...

The role of X-chromosome inactivation in female predispositionto autoimmunity

We propose that the phenomenon of X-chromosome inactivation in females may constitute a risk factor for loss of T-cell tolerance; specifically that skewed X-chromosome inactivation in the thymus may lead to inadequate thymic deletion. Using a DNA methylation assay, we have examined the X-chromosome inactivation patterns in peripheral blood from normal females (n = 30), female patients with a va...
